摘要 Probe-vector molecule (I) for detection of a target nucleic acid contg. a base sequence of formula (II) consists of a linear partly single-stranded DNA molecule comprising a long strand contg. the sequence of formula (III) and a short strand contg. the sequence of formula (IV). For any k, X'k is the base complementary to Xk, and Y'k is the base complementary to Yk, the ends of the probe vector being complementary to sections of the target DNA; m,n=numbers sufficiently large that when the probe-vector is added to the target DNA under hybridising conditions, stable hybridisation will occur between ends of the probe-vector and the complementary section of the target DNA to form a circular hybrid; the probe-vector is circularised only if the particular target sequence is present and has hybridised to the probe-vector; regions Z1Z2Z3---Zp and Z1'Z2'Z3'---Z'p of the probe-vector strands contain a replicon and confer a detectable phenotype such that bacteria transformed by the hybrid are detectable and distinguishable from untransformed bacteria by virtue of exhibiting the phenotype.
