The invention relates to a process for assaying urea or urease in liquid media, in particular in biological fluids. According to the invention, the sample, containing the urea to be assayed or the urease to be assayed respectively, is mixed with a first reagent essentially consisting of an aqueous solution of a chemical compound which is stable in the said solution at least during the period of the assay, whose colour varies as a function of the pH, within the pH interval ranging from about 5.5 to about 9, the said solution containing, where appropriate, urease or urea respectively, the said urease or urea being, otherwise, subsequently added in the form of an aqueous solution and constituting, in this case, a second reagent, and the concentration of the urea or of the urease respectively determined is deduced from the difference between the measurements of the optical density of the sample at the same wavelength of the visible spectrum which are carried out before and after the hydrolysing action of the urease, this difference being compared to the result obtained, under the same conditions, with a standard solution of urea or urease respectively.
