PURPOSE:To enable hydrolysis of polymer lignin at a high level by adding a lignin-related substance having phenolic hydroxyl groups, culturing an NK-1148 strain and using the resultant enzyme having about 88,000 molecular weight. CONSTITUTION:A culture medium containing a carbon source, such as glucose, a nitrogen source, such as peptone, salts, syringylglycerol-beta-syringyl ether, vanillyl alcohol, syringyl alcohol, etc., as a lignin-related substance is used to culture an NK-1148 strain (FERM P-9384) at about 28-33 deg.C. An enzyme- containing solution is then separated from the resultant culture to afford a crude enzyme by concentrating, salting out, precipitating, etc. Low-molecular impurities are subsequently removed therefrom to carry out purification by using chromatography. Thereby, three kinds of enzymes (F-1, F-2 and F-3) having properties of about 88,000 molecular weight and 2-2.5 isoelectric point are obtained. As a result, the polymer lignin can be converted into low molecular weight substances or decomposed without polymerization thereof. |
