PURPOSE:To separate extract of high concentration while controlling modification by extracting a specific component out of a sample by means of supercritical fluid, passing the supercritical fluid including extract through a column containing silica gel coated with silver nitrate and carrying out separation of extract. CONSTITUTION:Supercritical fluid of carbon dioxide and, if desired, an entrainer composed of ethanol are fed from a fluid feeding section 10. Then, extraction out of methyl ester of fish oil is carried out by means of supercritical fluid fed from the fluid feeding section 10 in an extraction section 12. Further, a separation section 14 is provided with a column 42 containing silica gel coated with silver nitrate through which the supercritical fluid containing said extract is passed for adsorption of the extract. Methyl eicosapentaenoate is separated by the supercritical fluid, and methyl decosapentanoate by the both supercritical fluid and entrainer in the column 42. As a result, extract of high concentration can be separated while controlling modification. |
