<p>A substitution phage vector μSK15 for construction of kDNA structure gene libraries along BamI and EcoRI restrictase identification sections has the size of 51.2 k.b.p of nucleotides, the vector capacity of 0.2 to 15.4 k.b.p of nucleotides. μSK15 consists of: DNA of phage μEMBL3; DNA of plasmid SK2ΔSR; a kanamycin resistant gene of plasmid pUC4K; Hind III-fragment of DNA phage μ of a size of 2.0 k.b.p of nucleotides. The host spectrum for the strain E. coli: LE392 = F', hsd R514 (rk, mk), sup E44, sup F58, lac YIOZ (lac IZY)6, galK2, galT22, metB1, trpR55, j.M.109 = recΔI, ΔIac pro, end A1, gyr A96, thi-1, hsd R17, sup. E44, rel A1, F', tra D36, pro AB?+, lac Iq¿ZΔM15. Q359 = hsdR¿k?-, hsd Mk+, sup F, Ø 80, P2. The claimed vector was deposited on 21.07.89 at the all-Union Collection of Industrial Microorganisms of the USSR Research Institute for Genetics and Industrial Microorganism Breeding and was registered under number PH No 688.</p> |
