PURPOSE:To easily obtain an establishing gene useful for the technological field for the genetic engineering, by finally introducing a DNA fragment composed solely of intron sequence into a prescribed gene originated from yeast, etc. CONSTITUTION:A yeast URA3 gene (A) is prepared by the genetic engineering of a yeast (S. cerevisiae). The component A is cut with a restriction enzyme forming cohesive ends. One of the ends is converted to a smooth end with a DNA polymerase and the other is converted to a smooth end with a single- chain specific DNAase to obtain a treated gene (B). A yeast-originated intron of a gene coding a polypeptide having a base sequence of formula (N and N' are complimentary with each other and are selected from A, C, G and T or their combination) is inserted in the right direction into the component B to produce an establishing gene. |
