摘要 |
PURPOSE:To produce a heat-resistant alpha-amylase in high efficiency, by synthesizing a recombinant plasmid obtained by integrating a DNA fragment having a specific heat-resistant amylase gene into a manifestation vector DNA. CONSTITUTION:A DNA fragment having heat-resistant amylase gene originated from Dictyoglomus thermophilum H-5-12 strain (FERM P No.7114) is integrated into a manifestation vector DNA to synthesize a recombinant plasmid. In the above process, Escherichia coli C-600 is transformed with a recombinant plasmid pDA201 containing an amylase gene originated from a 5.6KbP fragment of a chromosome DNA decomposed with Sau3AI among the genes. The culture of the transformant can be carried out aerobically at normal culture temperature. The objective heat-resistant amylase is separated from the cultured bacterial cell or liquid. |
