PURPOSE:To enlarge DNA content, cell size and catalase specific activity, by mutually subjecting auxotrophic stains induced from Candida SP-N16 to cell fusion and synthesizing a microorganism having high enzyme specific activity. CONSTITUTION:Candida SP-N16 (FERM P-425) is subjected to ultraviolet ray irradiation and reaction with a chemical varying agent such as nitroguanidine to form auxotrophic strain. The resultant auxotrophic strain is treated with mercaptoethanol to protoprastate the strain and the protoplastated auxotrophic strains are mutually subjected to cell fusion e.g. using polyethylene glycol. Then the aimed fused strain is separated utilizing complementarity of auxotrophic properties. A microorganism which is the resultant fused strain has each large DNA content, catalase activity and cell size compared with Candida SP-N16. |
