CLONED DNA OF HUMAN MOTILIN PRECURSOR, ITS FRAGMENT, PREPARATION THEREOF AND PLASMID HAVING SAID CLONED DNA OR FRAGMENT INTEGRATED THEREIN

PURPOSE:To mass-produce a motilin precursor, etc., by forming a cloned single- or double-stranded DNA containing about 550 nucleotides coding an amino acid sequence of a human motilin precursor. CONSTITUTION:An RNA derived from the human superior small intestine is converted into poly(A)RNA, which is used together with a vector primer DNA to prepare a cDNA library and transform a strain of Escherichia coli. On the other hand, the N-terminals of a mixture of 24 kinds consisting of 23 deoxyribonucleotides are labeled and used as a probe to screen the above- mentioned transformant cell by hybridization. Thereby a cloned double-stranded DNA is obtained as a hybridizable positive clone. The resultant DNA, as necessary, is decomposed into a single-stranded DNA by a conventional method.
