PURPOSE:To efficiently produce endo-alpha-N-acetylgalactosaminidase, by cultivating a microorganism, belonging to the genus Alcaligenes and having the ability to produce the endo-alpha-N-acetylgalactosaminidase. CONSTITUTION:A microorganism, e.g. F-1906 strain separated from soil, belonging to the genus Alcaligenes and having the ability to produce endo-alpha-N- acetylgalactosaminidase is cultivated. After completing the cultivation, microbial cells are separated from the resultant culture fluid by a means, such as centrifugation, and the cultivation supernatant liquid is fractionated with ammonium sulfate and then chromatography, such as ion exchange, gel filtration or affinity, is applied to afford the aimed purified endo-alpha-N- acetylgalactosaminidase. A culture fluid containing about 0.1-10% porcine gastric mucosal mucin treated with 1N sulfuric acid at about 80 deg.C for about 1hr is preferred as the culture fluid. |
