发明名称 PRODUCTION OF NOVEL PLASMID, MICROBIAL CELL AND POLYPEPTIDE HAVING ANTITUMOR ACTIVITY
摘要 PURPOSE:To provide a plasmid containing a DNA region coding an antitumor- active polypeptide and a DNA region imparting a host cell with an action to promote the extracellular excretion and useful for the production of said antitumor-active polypeptide by extracellular excretion. CONSTITUTION:The objective plasmid contains (A) a DNA fragment having a DNA region coding an antitumor-active polypeptide, a DNA region having promoter function to perform the manifestation control of said polypeptide and a DNA region coding a signal peptide and (B) a DNA fragment having a DNA region to impart a host cell with an action to promote extracellular excretion (preferably originated from plasmid pMB9) and a DNA region having promoter function. The DNA region having promoter function and the DNA region coding a signal peptide are preferably those originated from chromosome DNA of alkaliphilic Bacillus No.170 strain. A recombinant microorganism transformed with said plasmid is cultured and the antitumoractive polypeptide excreted from the microbial cell is collected.
