PURPOSE:To obtain a novel cDNA probe, containing a specific base sequence, capable of simply and readily judging the type of a human leukemic antigen (HLA class II DP antigen) and useful in the field of organ transplantation, etc. CONSTITUTION:All the RNA fractions are collected from a human-derived AKIBA cell which is a homozygote of DP antigen and subjected to fractionation treatment to provide a desired mRNA fraction, which is then treated with a reverse transcriptase to synthesize a cDNA. The resultant cDNA is subsequently converted into a double-stranded cDNA with a polymerase, treated with a nuclease and subjected to dC-tailing to afford a treated cDNA (A), which is then linked to a plasmid pAT153 (B) treated with a restriction enzyme (e.g. PstI) to provide a cDNA inserted plasmid (C). Escherichia coli x1776 is subsequently transformed with the component (C) to afford a transformant strain and a plasmid is separated from the obtained strain to provide a cDNA probe containing beta-chain gene of DP antigen containing part or all of the base sequence expressed by the formula, etc. |
