PURPOSE:To readily and efficiently obtain an exogenote product useful in cultivation engineering field in a high concentration, by cultivating specific Escherichia coli while controlling saccharide concentration of a culture medium and controlling expression of the exogenote. CONSTITUTION:A recombinant plasmid pSYPL-3 prepared by inserting L- phenylalanine ammonia-lyase (PAL) structural gene into a vector, etc., containing a gene capable of coding ampicillin tolerance to provide an Escherichia coli MT 10424 strain capable of expressing PAL. The resultant strain is then inoculated into a culture medium containing tryptone, etc., and cultivated at 40-45 deg.C cultivation temperature in the first process, control expression of the exogenote and multiply the amount of microbial cells. The cultivation temperature is then kept at 30-38 deg.C to carry out cultivation in the second process and control of exogenote expression is released to produce the aimed PAL which is an exogenote product in the culture medium. |
