<p>PURPOSE:To obtain the titled antibody, produced by a hybridoma prepared using an outer membrane protein C (OmpC) of Escherichia coli as an immunologi cal source and capable of directly detecting Gram-negative bacilli mixed in a sample, e.g. biological sample or food, etc., without pretreatment by heating. CONSTITUTION:A blend prepared by blending OmpC or a crude outer membrane fraction of Escherichia coli containing the OmpC with complete or incomplete Freund's adjuvant is injected into an animal, e.g. mouse, etc., to carry out sensitization and splenic cells or lymphocytic B cells extracted from the immunized animal are fused to myelomatous cells. The aimed specific antibody-producing cells are selected from the fused cells and cloned to provide a hybridoma capable of producing the aimed antibody. The above-mentioned antibody is cultivated in a normal culture medium for animal cells and reacted with OmpC trimer to afford a monoclonal antibody unreactive with the OmpC monomer and OmpC trimer. The afore-mentioned antibody is capable of linking to live cells of Escherichia coli and Citrobacter freundii.</p> |
