PURPOSE:To simply determine a base sequence with high accuracy, by a method wherein a unidimensional wave-form consisting of the position along the development direction of the separated and developed rows of bases of nucleic acid and a signal level is divided into two to smoothen the signal level to determine the base sequence. CONSTITUTION:The mixture of base specific fragments of nucleic acid labelled with a radioactive marker as a specimen is separated and developed according to various separation/development methods using a support medium. An autoradiograph is calculated by a radiation converting method and the digital signal thereof is obtained through a reading system. The digital signal of the unidimensional wave-form corresponding to four kinds of bases of DNA or RNA is successively changed over by an input/output change-over part 11 to be inputted to a smoothing operation part 12. A coefficient control part 13 sends a coefficient output signal to a coefficient table 14 at each time when a predetermined number of signals are inputted to the operation part 12. The operation part 12 receives the coefficient from the table 14 to perform smoothing operation. The determination of a base sequence is performed from the smoothed unidimensional wave-form.
