PURPOSE:A shuttle vector that contains individual genes from yeast and Escherichea coli, bears a specific regulatory area in the yeast and loses a part of genes from the Escherichea coli at the downstream of the regulatory area, thus being used as a vaccine material, because of its high manifestation efficiency. CONSTITUTION:A plasmid from Yeast S288CDNA is treated with a restriction enzyme to obtain a plasmid pAM82 (Figure I) which contains yeast genes such as arsI and genes from Escherichea coli such as pBR322 and consists of a polypeptide of 60,000 dalton (p60) constituting phosphatase and responds to the regulatory area (A) manifesting inhibitory acidic phosphatase trait in the yeast. The plasmid is treated with a restriction enzyme such as PvuII to add the BamHI linker to the site so that the plasmid is made cyclic again. Finally, about 1.4Kbp sequence from salI (651) in pBR322 to PvuII (2066) is removed from the downstream of the component A to give a shuttle vector such as pONYI (Figure II). |
