PURPOSE:To make it possible to use effectively a hybrid plasmid as a shuttle vector, by preparing the hybrid from a plasmid derived from Gram-negative bacteria and a plasmid derived from the genus Erwinia. CONSTITUTION:A hybrid plasmid prepared by subjecting a plasmid derived from Gram-negative bacteria and a plasmid derived from the genus Erwinia to restriction enzyme treatment by a restriction enzyme selected to give respectively a DNA fragment containing a replication starting point by the conventional method, reconstructing a plasmid according to, e.g. the Birnboim method, etc., transforming Escherichia coli, etc., having the transformation ability with the resultant plasmid, screening the transformant in a suitable culture medium to select a colony containing the aimed plasmid, cultivating the bacteria in the colony and obtaining the aimed hybrid plasmid according to the conventional method. |
