PURPOSE:To determine 3alpha-hydroxysteroid in high color sensitivity accurately, by using both a formazan and a formazan from a specific enzyme coupling system. CONSTITUTION:A test specimen is treated with 3alpha-hydroxysteroid dehydrogenase in the presence of nicotinamide adenine dinucleotide. Then, the prepared reaction product is treated with a tetrazolium or its salt, an electron carrier substance, and 3-oxosteroid-DELTA<1>-dehydrogenase or 3-oxosteroid-DELTA<4>-dehydrogenase or both, and a formed formazan is measured, to determine 3alpha-hydroxysteroid. Any of diaphorase, 1-methoxy-5-methylphenazinium methylsulfate, 9-dimethylaminobenzo- phenazoxonium chloride is used as the electron carrier substance.
