PURPOSE:To determine an enzymatic activity by rate assay process preventing the influence of the colored ingredient in the specimen, by the colorimetry of a phenylenediamine derivative using a synthetic substrate capable of isolating the phenylenediamine derivative by enzymatic reaction. CONSTITUTION:A synthetic substrate capable of isolating a phenylenediamine derivative by the active of the objective enzyme in the living body specimen such as serum, plasma, etc., is used as the substrate of the present process. The coupling product of the phenylenediamine derivative and an aniline derivative and having an absorption peak at >=700nm wavelength and a molecular extinction coefficient of >=40,000, is determined by colorimetry. Bilirubin oxidase fee from the inhibiting action to the objective enzyme is used as the coupling agent (condensation agent).
