<p>2-Amino 9-((1,3-dihydroxy 2-propoxy)methyl 1,9-dihydro 6H-purin-6-one is made by (a) bubbling dry HCl gas into a mixt. of 1,3-di-o-benzyl glycerol and paraformaldehyde in CH2Cl2 at 0 deg.C to give 2-o-chloromethyl 1,3-di-o-benzyl glycerol; (b) treating trimethyl chloro-silane with guanine at 40-50 deg.C in presence of triethylamine for 4 days to give tris(trimethyl silyl) guanine; (c) condensing these two prods. in diphenyl ether at 180-195 deg.C; (d) treating the prod. with methanol to give 2-amino 9-((1,3-di-o-benzyl 2-propoxy)methyl) 1,9-dihydro 6H-purin-6-one; and (e) hydrogenolysing with H2 in acid ethanol soln. in presence of 10% Pd on C.</p> |
