PURPOSE:To provide a transcription-promoting sequence obtained from the human virus, and useful especially for the manifestation of physiologically active substance such as gamma-interferon, interleukin II, etc. utilizing an eucaryotic cell. CONSTITUTION:The objective transcription-promoting sequence is produced from the human BK virus (e.g. wild strain WT501, mutant strain Pm411, Pm525, etc.) by incising the virus DNA with a restriction enzyme, Hind III. Concretely, the virus DNA is extracted and separated from a human BK virus, e.g. the mutant Pm525, and incised with the restriction enzyme, Hind III. Each fraction is separated and linked with TK gene. The TK gene is introduced as a compound plasmid DNA into a eucaryotic cell by calcium phosphate method, and the amount of manifestation is determined by counting the number of colonies formed in the HAT medium. The transcription-promoting activity can be found in the fraction C in the fractions A, B, C and D formed by the incision with Hind III. The base sequence of the fraction C is shown in the figure. |
