PURPOSE:To measure the autoantibody for an antigen by culturing primarily the hepato-cells with a plate for tissue culture, immobilizing the monolayer of the hepato-cells and bringing the serum to be inspected and a labeled effecting agent into immnoreaction on said plate. CONSTITUTION:Hepato-cells are cultured in the well of a plate for tissue culture, for example, Williams E medium. The primary monolayer of the hepato-cells in the well is then fixed by a fixing agent, for example, parafomaldehyde. The serum to be inspected and a labeled effecting agent are added onto the plate and the immobilized hepato-cell antigen and serum are conjugated by immunoreaction and the labeled effecting agent is conjugated with such conjugate. The conjugated quantity is then measured. A radioactive material, enzyme active material, etc. are used for the labeled effecting agent. The quick measurement of the autoantibody for the hepato-cell membrane antigen with good accuracy is thus made possible by making the cultured and immobilized hepato-cells on the plate and bringing the same into immunoreaction, by which the liver disease is adequately diagnosed. |
