PURPOSE:To obtain a novel amylase having improved properties to produce maltopentaose, by cultivating Bacillus cereus. CONSTITUTION:Novel Bacillus cereus (FERM-6648) is separated from soil. When this mold is aerobically molded, a novel amylase capable of producing maltopentaose is prepared and accumulated in the outside of the mold. 0.25ml of this enzyme is treated with 0.5ml of 1wt% soluble starch and 0.25ml of 50mM acetic acid buffer solution, 38wt% maltopentaose is formed from the starch after 5min, and 67wt% maltopentaose after 2hr. This enzyme acts on soluble starch, amylopectin, amylose, dextrin, and maltohexaose, but not on pullulan, dextran, and beta-cyclodextrin. |
