PURPOSE:To provide a gene coding a polypeptide having interleukin 2 (IL-2) activity, useful for the microbial production of IL-2, and prepared from mRNA separated from the cell originated from lymphocyte and capable of producing IL-2. CONSTITUTION:The cell originated from human lymphocyte and capable of producting IL-2 is cultured (preferably for a time to activate the lymphocyte and to initiate the poduction of IL-2 in the supernatant liquid of the culture medium), and the obtained cells are partially or completely disintegrated and solubilized. The product is fractionated by sucrose density-gradient centrifugation to obtain mRNA corresponding to IL-2 as 11-12S fractions. Complimentary DNA is synthesized from the obtained mRNA, connected with the replicon originated from microorganism, and introduced and amplified in a host microorganism. The clone having DNA capable of manifesting IL-2 polypeptide i separated, and the DNA inserted in the plasmid contained in said clone is separated to obtain the objective gene containing the fragment coding the polypeptide of formula. |
