PURPOSE:To obtain exactly a reagent capable of determining bilirubin concn. by incorporating a dye together with a promotor, a diazotizing agent, and hydrochloric acid so as to render light absorbance of a principal wavelength higher than that of a subsidiary wavelength up to a specified range and obtaining the difference between both light absorbances of them. CONSTITUTION:A red or blue dye, such as amaranth or Evans blue, not hindering the reaction between a reagent and bilirubin and being intrinsically stable against fading is added to the bilirubin determining reagent in such a concn. that the light absorbance at the principal determination wavelength near 550nm may be made higher than that of a subsidiary wavelength (longer in wavelength than the principal one where little absorbance is found in the sample serum, etc. such as 660nm or 700nm) by 0.01-1 per 10mm. light path. The determination reagent contains a reaction promotor, such as dimethylsulfoxide, a diazotizing agent, such as p-sulfobenzenediazonium-1,5-naphthalenedisulfonate, and HCl for completing the diazotization reaction. The use of this reagent enables exact determination of bilirubin concn., even if low, without causing blur, double peaks, etc. |
