PURPOSE:To enable the full-automatic analysis without generating problems such as the clogging of an arranged pipe or the like, by a method wherein a nitrogenous reducing agent and a ninhydrine solution with a new composition are used while a specimen to be analyzed is added to a separation buffer solution and said reducing agent is mixed in the liquid separated and eluted from a column and the ninhydrine solution is succeedingly mixed therein. CONSTITUTION:The specimen from an automatic sampler 9 is sent to an injector 10 and mixed with a separation buffer solution sent by a change-over valve 5 and a pump 6 while the resulting mixture is sent to a column 11. The column elution liquid to which a nitrogenous reducing is added is passed through a mixing pipe 12 while a ninhydrine solution containing no reducing agent is sent to be passed through a mixing pipe 13 and color development reaction is performed in a reaction tank 17. In this case, KCl and K2HPO4 are used in the separation buffer solution of amino-glucoside antibiotic. Analysis is performed in a hermetically full-closed system while all operations are controlled by a control part 16 and it is not necessitated that the solution is preserved in a light blocked state and N2-gas is flowed through the entire system. |
