PURPOSE:A DNA, containing a polynucleotide capable of coding the H-chain polypeptide in a human immunoglobulin (IgE), and usable for mass producing the H-chain polypeptide of the human IgE by utilizing the gentic manipulation technique. CONSTITUTION:A novel DNA, obtained by synthesizing a single-stranded cDNA with an mRNA capable of coding a human immunoglobulin (IgE) extracted and separated from an established human myelomatous cell U266 having the ability to produce the human IgE as a template by using a reverse transcriptase, converting the cDNA into a double-stranded DNA, digesting the resultant DNA with an enzyme (an exonuclease or endonuclease), adding an adapter to the digested DNA, introducing the resultant adduct into a plasmid, integrating the resultant plasmid into Escherichia coli, etc., cultivating the integrated Escherichia coli, and isolating the plasmid containing the DNA to give a double-stranded DNA capable of coding the H-chain polypeptide in the human IgE, and usable for producing the H-chain polypeptide in the human IgE by transforming a host cell, e.g. Escherichia coli or Bacillus subtilis, and cultivating the above-mentioned transformant cell. |
