Prodn. of several monoclonal anti-CALLA (common acute lympho -blastic leukaemia antigen) antibodies (Ab) comprises first fusing a Louvain rat myeloma cell line with spleen cells from a Louvain rat immunised with cells from a leukaemia patient. The hybrids are selected and isolated, and the resulting clones developed in Louvain rats and the Ab isolated from the resulting ascites. Pref. the myeloma line used is deficient in hydroxanthine guanine phosphoribosyl transferase and does not secrete immunoglobulins. The monoclonal Ab designated AL2, 3, 4 and 5 are new. Several Ab can be prepd. from a single cell fusion. The various Ab recognise all the membrane antigens of human lymphoid cells and are useful in diagnosis and oncohaematology. They are also potentially valuable in bone marrow transplant therapy of acute lymphoblastic leukaemia.
|
