PURPOSE:To improve solubility to aq. solns. of neutral regions and sensitivity of immunoassay by binding a compd. having a dissociating group or polar group to an antibody and changing the isoelectric point of the antibody. CONSTITUTION:Diazonium benzenensulfonate is easily obtained by diazotization of sulfanilic acid. When unreacted matter is removed by dialysis and an isoelectric point is measured by using a primary electrophoresis method, 6-7 PI before the reaction changes to about 5 PI after the reaction. The solubility near neutral pH is improved as well. A marking reagent is prepd. by using this chemical conversion method. Luminol is used for the marked compd. and alubmin as its carrier. When the quantity of the light emitted by the luminol is compared between the case in which amino groups are protected by the diazotized sulfanilic acid against antihuman IgG and the case in which no protection is given, the former exhibits intensity of light emission/1% protein= 800, while the latter exhibits 44.5, whereby the marked antibody of high solubility and high intensity of light emission is obtained. |
