Plasmogen is isolated by adsorbing plasminogen from plasma or serum or a plasminogen contg. plasma or serum fraction on to a water-insol. carrier contg. an amino-acid having an amino group in the epsilon-position to the -CO2H group, removing non-specific accompanying proteins by washing with an aqs. buffer soln. of pH 5.5 - 6.5 and 0.05-0.15 M, and then with a 0.05-0.15 Buffer soln. of pH 2.0-3.5 or 9.0-10.2, which also contains an amino-acid of above type in a molarity of 0.05-0.1, eluting and opt. enriching by precipitating with (NH4)2SO4, then concentrating e.g. by dialysis, and finally drying lyophilically. |
