NEW MATERIAL:A duplex polydeoxyribonucleotide containing a structural gene exhibiting 27-desamidosecretin. USE:A plasmid for massproducing inexpensively and quickly 27-desamidosecretin through genetic engineering, exhibiting a promoting action of the secretion of pencreatic juice by intravenous injection. PROCESS:A gene of base arrangement comprising a bond of the blocks A, B, and C is designed, each fragment S-1-S-16 is synthesized by a triester method preferably by solid phase method, purified and isolated by a reversed phase column. The 5'-hydroxyl group of each fragment is phosphorylated, bonded successively with DNA ligase in a buffer solution to form the blocks A, B, and C, and these are linked to give a structural gene. This gene is integrated in a vector plasmid such as the recognition site PstI of pBR322. |
