PURPOSE:To determine antigen in a specimen quickly and readily, by using an enzyme- labelled antigen of clenbuterols, an antibody obtained by injecting into an animal a haptenic antigen prepared by covalently bonding clenbuterols and a macromolecular compound, and a carrier containing a second antibody adsorbed thereon. CONSTITUTION:Clenbuterol (a medicine having a bronchodilatory action) of the formula (where R1 and R2 represent each chlorine or trifluoromethyl and X is -N2<+>) is covalently bonded with an enzyme such as beta-D-galactosidase through a group X to prepare an enzyme-labelled antigen. A haptenic antigen in which clenbuterol is covolently bonded with serum albumin (human or bovine) through a group X is injected into an animal to prepare a haptenic antibody (antiserum). A second antibody obtained by injection IgG of said animal into another animal ia fixed on a carrier of polystylene or the like to prepare a second antibody. Then, a reaction is caused between a specimen (plasma from a patient) and the enzyme-labelled antigen and between the haptenic antibody and the second antibody carrier. Thereafter, an enzymatic activity is measured for the labelled antigen bonded or non-bonded with the second antibody carrier and the level of the antigen in the specimen is measured with a high accuracy from a calibration curve prepared beforehand. |
