PURPOSE:To obtain a factor for suppressing proliferation of fibroblast cell and useful for the prevention and remedy of various diseases, by culturing a human diploid fibroblast cell and recovering and separating the factor from the supernatant of the cultured product. CONSTITUTION:A human diploid fibroblast cell, preferably FC cell which is a diploid fibroblast cell originated from human placenta is cultured in Eagle's MEM or Dulbecco's MEM containing 0.1-15% of bovine fetus serum or calf serum at 37+ or -0.5 deg.C for about 1-15 days under normal pressure and a supernatant liquid is separated from the cultured product by conventional method such as centrifugal separation. The supernatant liquid is concentrated by ultrafiltration and fractionated and purified by gel-filtration, affinity chromatography, etc., to obtain the objective factor for suppressing the proliferation of fibroblast cell. The factor has a molecular weight of 102,000+ or -10,000 and is stable at pH 2.5-9. The factor is useful for the treatment of tumor or retinopathy to which remedying effect is expected from the suppression of arterialization and is useful also for the treatment of diseases caused by the abnormal proliferation of fibroblast cell (e.g. rheumatoid arthritis or arteriosclerosis). |
