PROBLEM TO BE SOLVED: To provide a method for enhancing the hydrolysis of a target RNA by a hammerhead ribozyme. SOLUTION: The method comprises making compound(s) having -OR group(s) (wherein, R is H or an alkyl), at least one selected from the group consisting of e.g. polyethylene glycol, ethylene glycol, dextran, ficoll, glycerol, 1,3-propanediol, 2-methoxyethanol and 1,2-dimethoxyethane, coexist with a hammerhead ribozyme in an aqueous solution and hydrolyzing a target RNA at a temperature e.g. higher than 37°C in a bivalent ion concentration of lower than 100 mM in the aqueous solution.
