The present invention relates to a method for preparing S-HGB ((S)-3-hydroxy-gamma-butyrolactone) using hydrolase, and more particularly to a method for preparing S-HGB in a high purity by hydrolyzing S-BBL ((S)-beta-benzoyloxy-gamma-butyrolactone) in the presence of hydrolase. According to the present invention, the S-HGB having an optical purity can be obtained in a high yield under simple process conditions without requiring reaction conditions of high pressure and high temperature or complex operating conditions by hydrolyzing S-BBL with hydrolase.
