The invention has disclosed a substrate capable of extremely enhancing the substrate-incising activity of hepatitis C virus NS3 protease when treated with the NS3 protease in the presence of a peptide originating in hepatitis C NS4 and an amino acid sequence appropriate as the cleavage site of a peptide substrate to be used in a system for assaying the activity of the NS3 protease depending on changes in the fluorescence intensity. The invention has developed an NS3 protease assay system usable in screening NS3 protease inhibitors, which enables rapid, convenient, highly sensitive and multiple processing.
