Provided is a method for isolating a housekeeping gene LsTUA6 from genomic DNA derived from gourd, by using a pair of primers in a sequence list 1 and a sequence list 2. According to the present invention, by providing the method for isolating tubulin (LsTUA6) which is a housekeeping gene derived from gourd used as an understock when packaging watermelons, comparison and analysis on the level of expression of genes to figure out for each situation and tissues by using the housekeeping gene.
