PROBLEM TO BE SOLVED: To provide a method for detecting a mutation (T341C mutation) at the 341st position of a cDNA of an NAT2 (N-acetyltransferase 2) gene.
SOLUTION: A nucleic acid probe is obtained by amplifying a region containing the T341C mutation according to a PCR (polymerase chain reaction) and labeling the terminus with a fluorescent dye. The nucleic acid probe reduces the fluorescence of the fluorescent dye when hybridized. A melting curve analysis is carried out by measuring the fluorescence of the fluorescent dye with the nucleic acid probe composed of a specific base sequence. Thereby, the mutation is detected on the basis of the results in the melting curve analysis.
COPYRIGHT: (C)2006,JPO&NCIPI |
