NEW MATERIAL:A novel nuclease B-1 which attacks the single chain scission area of one of the DNA chains of a double-stranded deoxyribonucleic acid, and specifically splits the other DNA chain at its complementary area. USE:Extremely usefrl for separating a DNA fraction having specific genetic information from a genetic DNA. PROCESS:Strain belonging to Bacillus genus and capable of producing neclease B-1, e.g. Bacillus subtilis Marburg 168GSY 1026 (FERM-P. No. 4216) is inoculated in a medium containing decomposed protein, and cultured under agitation and aeration. The strain at its stationary growth stage is separated, and purified by lysozyme-treatment, ultrasonic disintegration, and/or ion exchange resin chromatography. |
