<p>Horizontally mounted spiral tube is used for preparative, isoelectric focussing of dissolved or suspended solids. The coil has many loops with the same dia. e.g. 38. It is made from an electrically non-conducting but good heat conducting material which is rigid and chemically resistant e.g. glass. Its axis is straight or in curves e.g. as a meander or zig-zag, lying in the horizontal plane. To each winding of the spiral a small tube is attached, pref. pointing upwards. The loops have as small a dia. as possible and the tube dia. remains uniform for the length of the coil. The i.d. of the tube is no greater than necessary to permit adequate dissipation of heat, e.g. 6-10mm. For use with isoelectric focussing techniques for separating high mol. wt. materials from a background of other materials using differences in their protolytic dissociation constant e.g. sepn. of albumen from ampholytes. The system described uses density differences to encouraging sepn. of the molecular types. Permits the sepn. of larger quantities than previously possible without other materials needing to enter the sepn.-space. The process is extended to materials of lower density than the soln. itself and to particle suspensions. For prepn. of vaccines and other proteins.</p> |
