<p>1325742 Sustained release compositions FARBWERKE HOECHST AG 15 Oct 1970 [17 Oct 1969] 49063/70 Heading A5B Sustained release pharmaceutical preparations which melt or soften above body temperature are made by (a) forming a melt comprising a pharmacologically active compound and a thermosoftening carrier in suitable proportions, (b) forming the melt into droplets, (c) cooling the droplets and (d) further processing the droplets without shaping them. The active compound may be phenylethyl-biguanide hydrochloride, 4-chloro-N-(2-furylmethyl) -5-sulphamoyl-anthranilic acid, or 1-phenyl-1- (2-pyridyl)-3-(dimethylamino) propane maleate. The carrier may be a glycol or polyglycol wax acid ester, a wax alcohol or a fatty acid amide. The melt may also include a surface active agent, e.g. polyoxyethylene stearate, polyoxyethylene sorbitan mono-oleate, or polyoxyethylene sorbitan monostearate, water-soluble inorganic compounds, polyvinyl pyrrolidone, and/or lecithine. It may be a eutectic mixture. The droplets of melt may be cooled in air, or in a liquid, e.g. a mixture of n-hexane, liquid paraffin and a proprietary refrigerant. The solidified droplets may be administered in capsules.</p> |
