Substances (A) that act on endothelin receptors (ETR) are identified by: (1) selecting a cell line that endogenously expresses ETR or preparing a cell line that stably expresses recombinant ETR; (2) transforming these cells with a recombinant vector comprising (a) a promoter (P) that is activated by an increase in intracellular second messenger concn. and (b) a reporter gene (RG) functionally linked to P; (3) establishing cells that transiently contain, or have stably integrated, the vector; (4) exposing selected cells to (A) and (5) measuring expression of RG.
