Universal virus-like particle (VLP) influenza vaccines

Described herein are influenza virus-like particles (VLPs) that display on truncated, re-engineered or remodeled HA molecules on their surface. Also described are methods of making and using these VLPs.

1. A virus-like particle (VLP) comprising
at least one influenza matrix protein; and an antigenic influenza HA polypeptide displayed on the surface of the VLP, the HA polypeptide comprising an influenza HA2 polypeptide having an extracellular domain, a transmembrane domain and a cytoplasmic domain, the extracellular domain of the influenza HA2 polypeptide comprising at least 173 amino acid residues, wherein the HA polypeptide is selected from the group consisting of: (i) an HA polypeptide comprising the influenza HA2 polypeptide and influenza HA1 polypeptides, wherein the influenza HA1 polypeptides consist of residues 17 to 23 and 241 to 346 of the HA1 polypeptide, as numbered relative to wild-type HA (SEQ ID NO:13); (ii) an HA polypeptide comprising the influenza HA2 polypeptide and influenza HA1 polypeptides, wherein the influenza HA1 polypeptides consist of residues 17-65 and 281 to 346 of the HA1 polypeptide, as numbered relative to wild-type HA1 (SEQ ID NO:13), and a heterologous linker between residues 65 and 281 of the HA1 polypeptides; and (iii) an HA polypeptide consisting of an HA2 polypeptide.
