PROBLEM TO BE SOLVED: To provide a fish muscle immersed medium, which prepares a large amount of a medium in a short time and slightly causes a change in property by culture, and to provide a vaccine against a fish disease, having a high vaccine effect using the same. SOLUTION: The medium is prepared by roughly cutting a fish muscle cut out from a fresh fish into about 1 cm square, feeding a proper amount of the obtained fish muscle to distilled water cooled to 4°C, keeping the fish muscle at 4°C for 24 hours and sterilizing and filtering the liquid by a filter and is a liquid. When a fish pathogenic bacterium is separated by using the medium, an isolated strain hardly having reduction in pathogenicity in comparison with a conventional medium is obtained. The fish pathogenic bacterium is inactivated to prepare a fish vaccine that prevents infection of cold-water disease having high possibility of infection of fishes and has a high vaccine effect of preventing infection of cold-water disease.
