PROBLEM TO BE SOLVED: To provide novel heat-resistant β-glucosidase showing hydrolysis activity having PNPG as a substrate at least under the condition of 100°C and pH5.5, and furthermore under the condition where extremely high-concentration glucose exists, polynucleotide coding the heat-resistant β-glucosidase, and a method for manufacturing cellulose decomposed product using the heat-resistant β-glucosidase.SOLUTION: Heat-resistant β-glucosidase having novel amino acid sequence is acquired by extracting DNA directly from hot spring high-temperature soil and performing a large-scale metagenome sequence of hardly culturable microbial flora. The enzyme is polypeptide which has hydrolysis activity having PNPG as a substrate at least under the condition of 100°C and pH5.5, and consists of a specific amino acid sequence, polypeptide which consists of an amino acid sequence one or several amino acids of which are deleted, substituted or added, or polypeptide which consists of an amino acid sequence having 80% or more of sequence identity with the specific amino acid sequence.SELECTED DRAWING: None |
