摘要 The inventors have surprisingly found that it is possible to increase the ratio of beta cells and progenitors thereof, obtained from undifferentiated cells by manipulation of culture conditions. In particular, they found that the use of hydrogen peroxide (H 2 O 2 ) at appropriate concentrations, in the culture medium of culture of embryonic pancreases containing stem cells, can significantly increase the ratio of beta cells obtained from stem cells.The present invention relates to an in vitro method for obtaining cells of the pancreatic endocrine lineage, and in particular of endocrine islets cells or precursor thereof, from undifferentiated cells, such as embryonic stem cells or induced pluripotent cells. The invention further encompasses cells of the pancreatic endocrine lineage obtainable by the present methods, compositions comprising said cells, and applications thereof.
