This disclosure relates to circular permutations of polypeptides and uses related thereto. In certain embodiments, the disclosure contemplates a library of catalysts produced by expression systems that translate primer amplified nucleic acids tandemly encoding portions of a protein of interest. In certain embodiments, the disclosure relates to methods of screening an enzmye library for catalytic activity on target starting materials. In certain embodiments, the disclosure relates to reduction reaction catalysts which are circular permutations of a cofactor dependent enzyme such as an old yellow enzyme and uses related thereto.
