PURPOSE: A transgenic Arabidopsis plant containing AtPP5 mutant gene construct is provided to enhance heat shock resistance and to enhance phosphatase activity. CONSTITUTION: A serine/threonine protein phosphatase 5(AtPP5) protein contains: three tetratricopeptide repeat(TPR) domain at amino acid residue 13-110 regions; a helix at amino acid 114-150 regions; a phosphatase active region at amino acid 190-459 regions; and a nuclear localization signal(NLS) region at amino acid 459-484 regions. The AtPP5 is denoted by sequence number 1. A method for producing a transgenic Arabidopsis plant which overexpresses AtPP5 protein or mutant thereof comprises: a step of assembling cDNA encoding serine/threonine protein phosphatase 5(AtPP5) or cDNA construct encoding a mutant of the protein; a step of inserting the cDNA into T-plasmid expression vector and co-culturing the vector with Argobacterium strain and Arabidopsis callus; a step of transducing the cDNA construct into Arabidopsis plant cells and transforming; and a step of growing the transformed plant cells.
