<p><P>PROBLEM TO BE SOLVED: To provide cyanine dye phosphoamidites for direct labeling of biomolecules, very stable and high in effects to cost, and to provide a convenient method for synthesizing phosphoamidites, requiring neither introduction step nor removal step of the protecting group. <P>SOLUTION: There is provided dye phosphoamidites, particularly phosphoamidites of a substituted crosslinked cyclic cyanine and related dyes, of general formula (I). In general formula (I), each dashed line represents carbon atoms necessary to form a fused substituted or unsubstituted ring; m is an integer of 1-18; Y and Z are independently selected from the group consisting of S, O, N, CH<SB>2</SB>and C(CH<SB>3</SB>)<SB>2</SB>; R<SB>1</SB>is an alkyl; (PAM) is a phosphoamidite group; X<SP>-</SP>is a negative ion; and Q is L-W, wherein L is a conjugated cyclic atom group and W is OR<SB>2</SB>, wherein OR<SB>2</SB>is a secondary alkyl.
