PROBLEM TO BE SOLVED: To provide a method, device, system and program for target nucleic acid measurement capable of accurately deciding the amount equivalent to the initial template amount in the cycle number of zero in an amplification reaction mixture using a theoretical expression to which the amount equivalent to the measured amplification amount of nucleic acid is applied as it is and which can accurately reflect an actual PCR amplification efficiency. SOLUTION: The amount equivalent to the initial template amount in an object to be examined is calculated by applying, to the theoretical expression, the thermal cycle number in a nucleic acid amplification reaction and the amount equivalent to the amplification amount of target nucleic acid measured every thermal cycle number. In this case, the theoretical expression includes: an environmental coefficient as an exponent parameter; a parameter of the amount equivalent to the initial template amount; and at least one parameter among an amount equivalent to a saturation amount upon the target nucleic acid amplification, a reaction acceleration coefficient, and a reaction inhibition coefficient.
